How to resuspend an oligo or primer
- Type the amount of oligo in the tube in nmol, from the tube label or the data sheet.
- Type the stock concentration you want, usually 100 µM.
- Add the volume of water or TE shown. To make a working solution, type its concentration and volume to see how much stock and water or TE to mix.
Frequently asked questions
What is the formula?
Volume in µL = nmol × 1,000 ÷ concentration in µM. For a 100 µM stock that is 10 µL per nmol.
Water or TE?
Low-EDTA TE (10 mM Tris, 0.1 mM EDTA, pH 8) keeps oligos stable longer in storage than water, and does not upset most PCR reactions at working dilutions.
